Agenda

Enhancing Antibody Discovery through in-silico methods at Bayer

Vanessa Verissimo, Senior Scientist
Bayer | Therapeutic Antibodies

In the field of antibody drug discovery, it is essential to accelerate the identification of therapeutic molecules while maintaining accuracy. This presentation delves into the shift from conventional high-throughput screening to in silico-assisted approaches, leveraging next-generation sequencing and machine learning to broaden antibody diversity and reduce reliance on wet-lab experiments. Bayer's advanced biologics platform enables the swift generation and purification of thousands of molecules, allowing for comprehensive multidimensional characterization. This innovative strategy fosters a quicker, more cost-effective early-stage drug discovery process.

Precision targeted therapies for neurodegenerative diseases

Susanne Snoek, Senior Scientist Vectorized Antibodies
VectorY Therapeutics | Research

Based in Amsterdam and founded in 2020, VectorY is dedicated to developing innovative vectorized antibody therapies for neurodegenerative diseases such as ALS, Huntington’s, and Parkinson’s.
By combining advanced antibody engineering with adeno-associated virus (AAV)-based delivery, our proprietary platform enables long-lasting, targeted treatments to the central nervous system.
This approach overcomes current therapeutic limitations, offering potential for disease modification and improved patient outcomes.

Validation of Structural-Based Machine Learning Designed In Vitro Libraries

Hoa Giang
Twist BioPharma | BioPharma Product Leader

At Twist Biopharma Solutions, we continue to develop and integrate new technologies to enable our antibody discovery partners to identify and optimize lead therapeutic candidates with speed, precision and accuracy. By synergizing Twist DNA synthesis with its library assembly technology and structural-based machine learning partnership, target specific antibody libraries were generated. Through successive rounds of panning, several highly potent and selective lead antibodies against C5aR were discovered. These antibodies exhibited functionally antagonistic effects on C5aR1-mediated signaling in cell-based assays.  In our second library, binders that could distinguish between two highly homologous undisclosed targets (~97%) are desired. Initial data suggests that these binders are present. This work demonstrates  how  the  combination  of  specific  DNA  synthesis  coupled  with informed design can be paired to successfully prosecute challenging targets for novel therapeutic discovery.

The dual face of IgG4 antibodies

Prof Theo (T.) Rispens, Principal Investigator
Sanquin | Antibody Structure & Function | Dept. Molecular Cell Bio. and Immuno. UMC

IgG4 antibodies are unique to humans. These antibodies are only a minor fraction of total IgG, but specific antibody responses can be dominated by this response, including autoantibody responses, and antidrug antibodies. Due to its special, non-inflammatory properties, it is often considered a 'tolerogenic' antibody, counteracting e.g. allergic IgE responses, but it may also cause autoimmune disease and concerns for immune evasion have been raised upon repeated vaccinations. Here we will discuss the special properties of IgG4 and the consequences for the associated (patho)physiological effects.

Immortalized patients’ derived B cell libraries for the interrogation and unbiased discovery of novel therapeutic targets and antibodies in oncology and beyond.

Casper Marsman, B Cell Platform Lead Department
Kling Biotherapeutics | B Cell Platform Development

Kling Biotherapeutics presents their B cell immortalization platform that transforms antibody discovery by capturing the complete diversity of patient-derived B cell repertoires. Their proprietary technology overcomes the limited lifespan of primary B cells, enabling comprehensive functional screening and immediate antibody production from culture supernatants.
The Kling B cell Tumor Libraries (TuLibs) approach captures B cells from all tissues and immunoglobulin isotypes, providing significant advantages over traditional BCR sequencing methods. Unlike labor-intensive recombinant antibody production, this platform allows direct functional screening while maintaining indefinite expansion capabilities. The immortalized B cells retain capacity for somatic hypermutation, enabling in vitro affinity maturation against specific targets. The Kling Bio technology has already enabled the discovery and development of two AML-specific antibodies targeting previously unknown tumor antigens, and identified cross-variant neutralizing antibodies against SARS-CoV-2.
Kling’s platform represents a paradigm shift in therapeutic antibody discovery, offering unprecedented access to natural human immune repertoires for developing targeted cancer and infectious disease therapies.

Carterra and Enpicom will be also presenting their solution:

  • Building blocks of an AI-Ready discovery Platform Nicola Bonzanni Founder and CEO, Enpicom
  • Carterra HT-SPR platforms: Essential lead discovery and screening Judicaël Parisot, Field Applications Science Team Lead Carterra
