Presentation Title: CBO-001 First-in-class L1CAM ADC

CBO-001 is CIS BIOPHARMA's lead antibody-drug conjugate targeting L1CAM in SCLC, one of the most aggressive lung cancers with limited treatment options and devastating patient outcomes. L1CAM is a cell adhesion molecule highly overexpressed in aggressive solid tumors, particularly neuroendocrine tumors. L1CAM expression correlates with tumor progression, metastasis, and poor prognosis. Its selective expression on cancer cells versus minimal presence in healthy tissues makes L1CAM an ideal target for precision oncology. The first-in-class L1CAM ADC CBO-001 combines a proprietary monoclonal antibody with state-of-the-art topoisomerase-I-inhibitor payloads delivered via cleavable linkers. Preclinical data demonstrate exceptional efficacy and safety, with superior performance compared to standard-of-care and emerging targets like B7-H3 and DLL3. CBO-001 is well on track for clinical development with scheduled clinical entry in 2027.

Presentation Title: Generative design of antibody Fc variants with synthetic and programmable effector function

Engineering antibody Fc domains with precisely tuned immune functions remains a key challenge in biologics development. We present FcGPT, a data-driven platform that integrates large-scale experimental Fc receptor binding data with predictive and generative machine-learning models to guide rational Fc design toward predefined functional objectives.

FcGPT learns quantitative relationships between Fc sequence features and binding to key receptors, including Fcγ receptors and FcRn, enabling systematic exploration of Fc sequence space. Embedded in a generative framework, the platform supports the design of Fc variants with tailored receptor engagement profiles, balancing inhibitory signaling and pharmacokinetic properties.

We demonstrate how FcGPT enables efficient Fc optimization, reduces experimental iteration, and provides a generalizable approach for developing next-generation antibody therapeutics requiring controlled immune modulation.

Presentation Title: AbDirect: Protein-Centric Discovery of High-Potency Human Antibodies from Patient Serum

While single-cell B-cell receptor sequencing (scBCR-seq) has proven successful for accessing B-cell repertoires, it does not directly capture genuine secreted, in vivo-matured antibodies present in human biofluids. AbDirect is a scalable protein-centric platform that enables standalone discovery and de novo sequencing of functional antibodies directly from small volumes of serum and other biofluids of elite responders, yielding serological leads with proven safety and efficacy in humans. In a COVID-19 benchmark using potent convalescent plasma from which an early-pandemic scBCR-seq study identified no neutralizers, AbDirect recovered 18 distinct clones with diverse reactivity profiles. Eight of these demonstrated neutralization, including multiple highly potent with IC50 <1.4 nM. This complementary approach accesses the immunologically distinct serological compartment and opens a powerful route to high-quality, pre-validated therapeutic antibody leads across oncology, autoimmunity, and infectious disease programs.

Presentation Title: Redefining High-Throughput SPR: The Carterra VegaTM Platform

Carterra® provides innovative technologies designed to accelerate the discovery of novel therapeutic candidates. Its high-throughput platforms—LSA, LSAXT, Ultra, and Vega - support monoclonal antibody (mAb) and small-molecule screening and characterization by combining patented microfluidics with real-time high-throughput surface plasmon resonance (HT-SPR) and advanced analysis and visualization software. Compared with traditional label-free platforms, these systems can deliver up to 100-fold higher throughput in a fraction of the time while using significantly less sample.

Through partnerships with industry leaders, Carterra is helping to develop high-throughput biological tools that support artificial intelligence (AI), machine learning (ML), synthetic biology, and in silico discovery workflows. By enabling rapid and comprehensive screening early in the discovery process, Carterra helps researchers identify unique epitopes and novel therapeutic candidates more efficiently—driving innovation, accelerating development timelines, and expanding intellectual property opportunities.
